|a Low-molecular-weight polyamines such as spermine and spermidine are ubiquitous in all forms of life and are involved in many cellular functions such as cell growth. Pathological altered polyamine concentration are associated with a variety of diseases. We thus aimed for their analysis from serum using reversed-phase liquid chromatography coupled to detection with quadrupole time-of-flight mass spectrometry. We tested 15 polyamines for their response, developed a multiplex target method, and refined it for five active substances including spermine and spermidine, which induced expression of the autophagy protein Atg8/LC3-II in a neuronal cell line.

|a Niedermolekulare Polyamine wie Spermin und Spermidin finden sich in allen Lebensformen und sind an vielen zellulären Funktionen wie dem Zellwachstum beteiligt. Veränderte Polyamin-Konzentrationen wurden in verschiedenen Krankheiten gefunden. Wir stellten uns daher das Ziel, sie aus Serum mittels Umkehrphasen-Flüssigchromatographie gekoppelt an Quadrupol-Flugzeit-Massenspektrometrie zu bestimmen. Wir untersuchten 15 Polyamine, entwickelten eine Multiplex-Methode und entwickelten diese besonders für die fünf aktiven Substanzen einschließlich Spermin und Spermidin, die die Bildung des Autophagy-Proteins ATG8/LC3-II in einer neuronalen Zelllinie induzierten.
