For research purposes, snake venom is often pooled to obtain sufficient amounts. However, considerable knowledge is necessary to identify snake species and utmost care is required to keep individually venoms well labeled during handling and venom pools clean. 1D-PAGE is an easy means as a first quality control to distinguish elapid from viper venoms. Moreover, the presence of significant amounts of three-finger toxins, which are hallmarks of elapid, not viperid, snakes, in proteomic results, e.g., from venoms of Echis species, indicates contamination of the venom pool. These small proteins can bet traced by mass spectrometry-based techniques. We provide examples for such experiments.
Zu Forschungszwecken wird Schlangengift oft gepoolt, um genügend Material zu erhalten. Es ist jedoch beträchtliches Fachwissen erforderlich, um verschiedene Schlangenspezies korrekt zu identifizieren, und große Sorgfalt ist nötig, um die individuellen Giftproben richtig zu handhaben und die Giftpools kontaminationsfrei zu halten. 1D-PAGE ist ein einfaches Mittel als erste Qualitätskontrolle, um Elapid- von Vipern-Gift zu unterscheiden. Zusätzlich indiziert die Anwesenheit von drei-Finger-Toxinen, die typisch für Elapids, aber nicht für Vipern sind, z.B. im Gift von Echis Spezies, eine Kontamination des Giftpools. Diese kleinen Proteine können massenspektrometrisch nachgewiesen werden. Wir zeigen Beispiele für solche Experimente.
